چکیده

A special feature of eggs of several species of annelids and molluscs is the formation of a polar lobe at stage-specific intervals during the cleavage phase. In the scaphopod Dentalium, a polar lobe develops at the first, the second and the third cleavage (Wilson, 1904). The cytoplasm set apart in the polar lobes is ultimately confined to the D blastomere and its derivative cells. Wilson observed that after removal of the polar lobe at the first cleavage a larva develops which lacks an apical tuft and the post-trochal region. After removal of the polar lobe at the second cleavage a larva develops which lacks most of the post-trochal region but possesses an apical tuft. A similar result to the one following the removal of the first polar lobe is obtained by removal of the vegetal one-third of an unfertilized egg. These results indicate that morphogenetic determinants present in the first polar lobe are already set aside in the uncleaved and unfertilized eggs, and that the determinants for the apical tuft are no longer present in the second polar lobe. The study of maturation in amphibian oocytes has shown that a relationship exists between nuclear DNA and Feulgen-staining granules appearing in the cortex after breakdown of the germinal vesicle (Brächet 1965, 1967). As Brächet pointed out this cortical localization may be significant with respect to the importance of the dorsal cortex (grey crescent) in later development. Therefore, we have studied the distribution of DNA in the egg cells of Dentalium as far as it can be traced with the Feulgen method.

The localization of morphogenetic factors in uncleaved eggs of Dentalium.

The effect of removing parts of unfertilized and fertilized eggs of Dentalium has been studied. Up to 70 % of the volume of a 1st polar lobe can be removed from the vegetal side of both unfertilized and fertilized eggs, without influencing apical tuft formation. The post-trochal region is reduced. These eggs form a reduced polar lobe at first cleavage. After removal of more than 70 % of the vol...

The Ultrastructure and Histochemistry of a Nematode-induced Giant Cell

The development of giant cells induced by the nematode Meloidogyne in tomato roots has been followed under controlled growth conditions and the ultrastructure and histochemistry of these structures have been examined. Entry of the nematode larvae into the roots took place within 24 hours; giant cell formation started on the 4th day and involved breakdown of the cell walls accompanied by thicken...

Detection of avian leukosis virus (ALV) in albumen of Shiraz commercial and local layer flocks using ELISA and RT-PCR

Avian leukosis viruses (ALVS) cause different types of tumours in poultry and can affect the health and egg production of the birds. To investigate the presence of the virus in chicken layer flocks in Shiraz, 222 egg albumen from local layer breeder (25 eggs), local layer grand parent (30 eggs), broiler breeder (60 eggs), commercial layer (46 eggs) and broiler grand parent (61 eggs) were tested...

Reporting an Experience: Improving the Feulgen Staining Technique for Better Visualizing of Nucleus

Among different staining methods used to demonstrate the nuclear abnormalities, Feulgen is one of the most reliable method. Feulgen staining is specific, sensitive method for evaluating the DNA damages. It has been shown that using non-DNA specific stains for monitoring the nuclear anomalies lead to false-positive or false-negative results. From self-experience, immersing the stained slides in h...

Increasing salt tolerance in Olive, Olea europaea L. plants by supplemental potassium nutrition involves changes in ion accumulation and anatomical attributes

The effects of supplemental potassium were studied on growth, ion concentration and anatomical parameters in one year old olive trees, Olea europaea L., grown in sand culture in greenhouse at different levels of NaCl for 80 days. The experiments were conducted in a completely randomized design as a factorial. Factor one was salinity (0, 40 or 80 mM NaCl) and the second factor was potassium leve...
